For methylation evaluation we have methylation activity assays, DNA purification kits, PCR enzymes and master mixes, methylation sensitive restriction enzymes, and basic tools for transfection. We offer Luciferase reporter assays you can use for detection of HDAC, SIRT, methyltransferase and various protease activities. You will also find epigenetics protein purification and interaction kits to analyze histone marks and identify protein binding partners rapidly and efficiently.

Epigenetics Research Basics

Epigenetics is the study of heritable changes in gene expression that arise from changes in chromosomes without alteration of DNA sequence. The coordination between DNA methylation and silencing histone marks leads to compaction of chromatin and gene repression.

The methylation status of a DNA sequence can be determined using a variety of techniques, such as restriction enzymes that are sensitive to methylation. Bisulfite sequencing is another way to assess DNA methylation. After bisulfite conversion and DNA cleanup, the DNA is amplified by whole genome PCR, and the amplified products are analyzed using one of several methods.The firefly luciferase reporter protein (Fluc) can also be used to assess DNA methylation.

To screen for potential histone deacetylase (HDAC) inhibitors, Promega offers the HDAC-Glo™ I/II and SIRT-Glo™ Assays to measure relative activities of HDAC class I and II enzymes and sirtuins, respectively. Promega also offers a number of kinase enzyme systems to monitor the activity or identify inhibitors of different kinases involved in histone phosphorylation.
