Quantify Targeted Cell Killing with Confidence
Principle of the HiBiT TCK Bioassay. Cytotoxic mAbs and/or effector cells are incubated with target cells expressing a HiBiT fusion protein. Upon killing of the target cell, the HiBiT fusion protein is released and binds extracellular LgBiT to create a functional NanoBiT® Luciferase enzyme. Luminescence is measured using a luciferase substrate and the GloMax® Discover System.
Applications
Antibody-Dependent Cell-Mediated Cytotoxicity (ADCC)
HiBiT TCK Bioassays reflect the ADCC MoA of biologic drugs and demonstrate specificity for cell surface antigens. H929 Cells (HT-HiBiT) were incubated with Human PBMC, ADCC-qualified effector cells in the presence of serial titrations of antibodies, as indicated. After a 5-hour induction, Bio-Glo-NB™ TCK Reagent was added and luminescence quantified using the GloMax® Discover System. Data were fitted to a four-parameter logistic curve using GraphPad Prism® software. HT = HaloTag, the fusion partner for the HiBiT tag.
T Cell-Dependent Cell-Mediated Cytotoxicity (TDCC)
HiBiT TCK Bioassays reflect the TDCC MoA of biologic drugs and demonstrate specificity for cell surface. H929 Cells (HT-HiBiT) were incubated with human T Cells (CD8+) in the presence of bispecific T cell engagers (BiTE®). After a 24-hour induction, Bio-Glo-NB™ TCK Reagent was added and luminescence quantified using the GloMax® Discover System. Data were fitted to a four-parameter logistic curve using GraphPad Prism® software. Data were generated using thaw-and-use cells. HT = HaloTag, the fusion partner for the HiBiT epitope.
CAR-T Cell-Mediated Cytotoxicity
HiBiT TCK Bioassays can be used in CAR-T applications. H929 Cells (HT-HiBiT) were incubated with serially diluted human BCMA CAR-T cells at the indicated E:T ratios. After a 4-, 24-, 48- or 72-hour induction, Bio-Glo-NB™ TCK Reagent was added and luminescence quantified using the GloMax® Discover System. Data were fitted to a four-parameter logistic curve using GraphPad Prism® software. HT = HaloTag, the fusion partner for the HiBiT tag.

When H929 (HT-HiBiT) target cells are cocultured with appropriate effector cells (e.g., CAR-T cells, PBMCs or cytotoxic T cells), they can be specifically used to monitor the activity of biologics directed against receptors expressed on the H929 Cells (HT-HiBiT), such as CD38 or BCMA. When H929 Cells (HT-HiBiT) are used in assays with biologics against TAAs they do not express, target cells are not lysed and no increase in luminescent signal is observed. Representative data for different MoA are above.

- 5.2. Use NanoBRET™-branded energy acceptors (e.g., NanoBRET™ tracers, NanoBRET™ dyes, BRET-optimized HaloTag® ligands) for all determinations of energy transfer activity by this material and its derivatives; or
- 5.3. Contact Promega to obtain a license for the use of the material and its derivatives for energy transfer assays to energy acceptors not manufactured by Promega. No license is needed if the energy transfer acceptor is a genetically encoded autofluorescent protein.

1. Usage and Transfer Restrictions:
- 1.1. Recipient may use this material for research use only, which includes but is not limited to drug discovery and development, quality assurance testing and product release assays. No transfer, resale or commercial use of this material is allowed without the written consent of Promega Corporation.
- 1.2. “Commercial use” means any and all uses of this material or derivatives by recipient for monetary or other consideration, including product manufacture, providing a service, information or data to unaffiliated third parties and resale of the material for any use.

2. Modification Restrictions:
- 2.1. Recipient shall have no right to modify, derivatize or otherwise create variations of the nucleotide sequence of any proprietary gene, including but not limited to luciferase, NanoBiT® technology (e.g., HiBiT), HaloTag® technology or genes stably transfected within the cells.
- 2.2. Recipient shall have no right to genetically engineer or otherwise modify the cell line.

3. Propagation and Storage:
- 3.1. The recipient may not propagate the cells, unless material is marked as Propagation Model, Cell Bank or Master Cell Bank; then recipient may propagate and store the cells for long-term use.
