Caspase-Glo® 3/7 Assays

The Caspase-Glo® 3/7 Assays detect caspase-3/7 activity of cells in culture to quantitate the amount of luminescence, which is proportional to the amount of caspase activity present, an indicator of apoptosis.

Jurkat cells were seeded in all-white 96-well plates from 10 cells/well to 100,000 cells/well (n=8 replicates). Cell viability was determined by adding an equal volume of CellTiter-Glo® or CellTiter-Glo® 2.0 Reagent to all wells, and the signal was read on a MyGlo™ Reagent Reader. These graphs were generated using Microsoft Excel.

Jurkat cells were seeded in all-white 96-well plates at 10,000 cells/well and treated with Bortezomib, Palbociclib or Anti-Fas (human, activating) clone CH11 for 24 hours. Data were fit using four-parameter logistic curves. Data were generated using the MyGlo™ Reagent Reader with the ProNect™ Data Platform.

Caspase-Glo® 3/7 Assay

Jurkat cells were seeded in an all-white 96-well plate from 10 cells/well to 100,000 cells/well and treated with vehicle or 50ng/ml anti-Fas (clone CH11; EMD Millipore Cat.# 05-201) to induce apoptosis (n = 4 replicates per condition). Caspase-3/7 activity was determined by adding an equal volume of Caspase-Glo® 3/7 Reagent to all wells. Assay signal was read on a MyGlo™ Reagent Reader (integration time = 1 second). These graphs were generated using Microsoft Excel.

Jurkat cells were seeded in an all-white 96-well plate at 10,000 cells/well and treated with increasing concentrations of anti-Fas (clone CH11) for 4.5 hours to induce apoptosis. Subsequently, Caspase-Glo® 3/7 Reagent was added, and data was generated using the MyGlo™ Reagent Reader with the ProNect™ Data Platform. Data were fit using four-parameter logistic regression.

If you need additional coverage during or after the MyGlo™ Reagent Reader standard warranty period is over, a MyGlo™ Reagent Reader Service Agreement is available. The MyGlo™ Reagent Reader Service Agreement covers all parts, labor and shipping to and from our authorized service locations. This Agreement does not include repair or replacement necessitated by accident, neglect, misuse, unauthorized repair or modification of the instrument.

If a problem arises with the instrument, please contact Promega Technical Service for support. E-mail: techserv@promega.com. If further action is required, you will be given repair options and assigned a return authorization number. Promega is not responsible for instrumentation returned without an authorization number.

To obtain service during the covered period, please take the following steps:

  1. Contact Promega Technical Services.
  2. Carry out tests as suggested by your Technical Services contact.

If it is determined that further action is required, you will be given options and assigned a return authorization number. Promega is not responsible for instrumentation returned without an authorization number.

Before returning the instrument, you will be responsible for cleaning it. If the instrument has been exposed to any chemical hazards, consult the Technical Manual for cleaning instructions.
