Tracking Cellular Protein Localization and Movement in Cells with a Flexible Fluorescent Labeling Technology

Summary

By knowing where a protein localizes in cells, researchers are one step closer to understanding a protein’s function. There are many methods for imaging proteins within cells including immunofluorescence-based assays and fluorescent protein tagging, but each of these methods limits the types of studies that can be performed. Here we describe the HaloTag® Technology, which is a small, universal protein tag engineered to covalently tether different functional groups to HaloTag® fusion proteins in living cells (or in vitro) such as, in the case of this webinar, a variety of fluorescent dyes. This technology enables researchers to easily perform many different types of protein imaging experiments including simple protein localization and the protein movement within a cell in real-time. This webinar will provide an overview of this unique technology including how it works and the various applications possible with HaloTag® with a focus on protein imaging. Then we will provide data illustrating these capabilities using both data generated in-house and in published, peer reviewed scientific publications.

Chad Zimprich is a Research Scientist in the Custom Assay Services (CAS) division of the Integrated Biology Group at Promega Corporation (Madison, WI). CAS is focused on gene delivery and imaging technologies, target engagement for small molecule drug discovery, and bioassay development for therapeutic antibodies. The focus of the research is both catalog and custom driven which has led to numerous commercialized assays. He has been awarded 14 U.S. Patents relating to the HaloTag® Technology and co-authored several peer reviewed journal articles.
